Adderall XR, with $111.4 million revenues in the first quarter of 2012, is one of the top revenue grossers at Shire. As per data provided by IMS Health National Prescription Audit, Adderall XR enjoys a 7% share of the US ADHD market. Apart from this, the company has two authorized generic partners - Teva Pharmaceuticals (TEVA) and Impax Laboratories (IPXL). We believe that the entry of additional generic versions of Adderall XR will eat into Adderall XR revenues.

Parents completed a modified version of the BSEQ during each week of the trial. Appetite, stomachaches, and insomnia were rated as worse by parents while children were receiving either dose of Adderall; headaches were rated as worse when children were receiving the higher dose of Adderall. Parents rated certain side effects, including staring/daydreaming, sadness, euphoria, and anxious/irritable, as worse during placebo regimens. Conclusions. We found that Adderall is highly efficacious in our population of youth diagnosed with ADHD. In addition, Adderall is well-tolerated with a side effect profile similar to that reported for other psychostimulants.

In the poor or nonresponder group of 9 patients (38%), all experienced side effects (including a more substantial decrease in appetite, gastrointestinal upset, migraine headache, and heart palpitations; see Table 2), prompting each one to discontinue the Adderall within the first 3 weeks of the trial. Of note, 4 of 7 patients in the study with a comorbid anxiety diagnosis experienced near-immediate symptoms of acute anxiety (including diaphoresis, tremor, shortness of breath, and an impending sense of doom) at the start of Adderall therapy.

Amphetamines have a high potential for abuse. Administration of amphetamines for prolonged periods of time may lead to drug dependence and must be avoided. Rapid improvements on teacher ratings and math performance were observed by 1.5 hours after administration, and these effects dissipated by the end of the day.
