Adderall XR, with $111.4 million revenues in the first quarter of 2012, is one of the top revenue grossers at Shire. As per data provided by IMS Health National Prescription Audit, Adderall XR enjoys a 7% share of the US ADHD market. Apart from this, the company has two authorized generic partners - Teva Pharmaceuticals (TEVA) and Impax Laboratories (IPXL). We believe that the entry of additional generic versions of Adderall XR will eat into Adderall XR revenues.

Parents completed a modified version of the BSEQ during each week of the trial. Appetite, stomachaches, and insomnia were rated as worse by parents while children were receiving either dose of Adderall; headaches were rated as worse when children were receiving the higher dose of Adderall. Parents rated certain side effects, including staring/daydreaming, sadness, euphoria, and anxious/irritable, as worse during placebo regimens. Conclusions. We found that Adderall is highly efficacious in our population of youth diagnosed with ADHD. In addition, Adderall is well-tolerated with a side effect profile similar to that reported for other psychostimulants.

Amphetamines have a high potential for abuse. Administration of amphetamines for prolonged periods of time may lead to drug dependence and must be avoided. Particular attention should be paid to the possibility of subjects obtaining amphetamines for nontherapeutic use or distribution to others, and the drugs should be prescribed or dispensed sparingly.

Adderall is composed of mixed amphetamine salts, which work by increasing the amount of the active neurotransmitters, norepinephrine and dopamine, in the brain. More formally known as Dextroamphetamine, it acts primarily on the DA systems. The primary reinforcing and behavioral-stimulant effects of amphetamine enhance dopaminergic activity, primarily in the mesolimbic dopaminergic pathway.

For those with ADHD, the drug increases alertness, libido and overall cognitive performance, improving mood and stemming fatigue. For the last 15 years, treatment with Adderall has helped significantly improve many lives. But for those who do not have the disorder, the drug, like other powerful amphetamines, causes euphoria, increasing the risk of addiction. According to the CDC, 1 in 12 Americans is diagnosed with ADHD and sales of Adderall have soared more than 30-fold since 2001.
